Typische Anwendungen für die Dunkelfeldmikroskopie

Das Dunkelfeld eignet sich als Kontrastverfahren immer dann besonders, wenn transparente Zellen oder Mikroorganismen sichtbar gemacht werden sollen, die selbst wenig Kontrast haben. Typische Beispiele sind zB Amöben oder Epithelzellen, zB von der Mundschleimhaut. Hier kann dann auf eine Fixierung und anschließene Färbung verzichtet werden. Dunkelfeldmikroskopie eignet sich auch besonders für Lebendkulturen von Wasserorganismen oder Algen, die man ja in Bewegung sehen möchte, was eine Färbung meistens ausschließt. Zusätzlich zum erzielten Kontrastgewinn sind Dunkelfeldbeobachtungen auch ästhetisch besonders reizvoll!

Schiefe Beleuchtung

Bei der schiefen Beleuchtung wird eine Kontrastierung erzielt, indem ein Teil bzw. eine Hälfte des normalen Beleuchtungsstrahlengangs abgedeckt wird. Kommerzielle Lösungen für dieses Kontrastverfahren sind leider selten, man kann diesen Effekt aber mit einer selbst angefertigten Blende / Schieber in der Filterebene des Kondensors erzielen. Oft kann auch ein vorhandener Filterhalter oder eine Hilfslinse am Kondensor teilweise in den Strahlengang eingeschwenkt werden. Der Rahmen des Filterhalters bzw. der Feldlinse blockiert dann einen Teil des Lichts. Das Präparat wird dann schräg durchstrahlt, es ergibt sich quasi ein Schattenwurf an Feinstrukturen des Präparats.

Phasenkontrast

Beim Phasenkontrastverfahren wird eine Kombination aus einem speziellen Kondensor mit wechselbaren Blenden (diese erzeugen einen Lichtring) und entsprechenden Phasenkontrastobjektiven (mit Phasenring) verwendet. Das Licht, welches das Präparat durchstrahlt, wird dort gebeugt und erfährt an Bestandteilen mit unterschiedlichem Brechungsindex eine Phasenverschiebung. Licht, welches keine Beugung erfährt, wird am Phasenring geblockt. 

Üblicherweise werden separate Phasenkontrastsätze angeboten. Eine ausschließliche Ausstattung des Mikroskops nur mit Phasenkontrastobjektiven ist in der Regel nicht empfehlenswert, weil zwar die Auflösung der Objektive durch den Phasenring nicht beeinträchtigt wird, der Kontrast aber schlechter ist als bei normalen Hellfeldobjektiven. Auch für Polarisation eignen sich diese Objektive nicht. 

Ein Phasenkontrastkondensor muss sorgfältig zentriert werden, damit Lichtring und Phasenring aufeinander abgebildet werden. Daher ist ein Wechsel zwischen Phasenkontrastkondensor und Hell-oder Dunkelfeldkondensor aufwändig. Man kann aber einzelne Hellfeldobjektive im Revolver neben mit Phasenobjektiven verwenden, denn der Phasenkontrastkondensor hat einen Durchlass für Hellfeld. Für die Zentrierung bzw. die Höheneinstellung des Kondensors ist im Phasenkontrastsatz ein Hilfsokular (auch "Einstellfernrohr" genannt) enthalten, so dass auf die Ebenen von Phasen- und Lichtring scharfgestellt werden kann.

Die Bedingungen für Phasenkontrast sind optimal nur für grünes Licht erfüllt. Daher ist ein entsprechendes Grünfilter hilfreich. Da ohnehin farblose Objekte im Phasenkontrast beobachtet werden, ist eine Einfärbung des Bildes tolerierbar.

Typische Anwendungen für Phasenkontrast

Ähnlich wie beim Dunkelfeld eignet sich Phasenkontrast für Objekte, die farblos sind und wenig Kontrast haben. Darunter fallen z.B. Planktonorganismen oder Belebtschlamm. Auch Zellkulturen oder Zellen im Urinsediment können mit Phasenkontrast besser dargestellt und damit auch schneller und sicherer beurteilt werden als im Hellfeld. Auch die Zählung von Asbestfasern erfolgt bei einigen Standardprotokollen im Phasenkontrast.

Polarisation

Durch Polarisation können optisch aktive  bzw. doppelbrechende Präparatstrukturen besonders hervorgehoben werden. Das können zB Minerale sein, aber auch viele Kunststoffe, Naturstoffe wie Stärke etc. zeigen diesen Effekt. Weniger bekannt ist, dass man zB auch an lebenden Organismen interessante Strukturen hervorheben kann, so zB Muskelfasern von Daphnien oder Rädertierchen. Die Polarisation dient in der Industrie vor allem zur Charakterisierung von Materialien. Ist die Schichtdicke eines Materials bekannt, so kann anhand der entstehenden Interferenzfarben auch auf das Material geschlossen werden. In der Materialforschung wird die Polarisation zB auch zur Untersuchung von Spannungen im Material durch Spannungsdoppelbrechung genutzt. Besonders Spritzgussteile oder Kunststofffolien bzw. Fasern können so auf Herstellungsfehler untersucht werden. Die klassische Anwendung ist natürlich die Geologie / Mineralogie, bei der Gesteinsdünnschliffe in polarisiertem Licht untersucht werden.

Aufgrund der Spektralverteilung sind in der Polarisationsmikroskopie Halogenbeleuchtungen besser geeignet als LED.

BRESSER Science MPO 401 Microscope
In contrast to usual light, polarized light has a preferred vibration plane of the light waves. Polarized light lets be seen anisotropic objects, whose observable properties are depending on the direction, "in a new light": crystals, but also animal hairs, feathers, muscles, nerve fibres and vegetable cell walls, which have a preferred direction of its biomolecules, show structures now, which were hidden in the normal bright field image. Images made by polarized light are often characterized by a fantastic coloring. Polarization microscopes are used particularly in mineralogy, geology, biology and medicine. In order to change the object's direction in a controlled way, the Science MPO 401 has a round, center-adjustable and full rotatable object stage with degree scale. The illumination and observation unit encloses e. g. the halogen bulb lamp (20 W) with dimmer, the field diaphragm (used to achieve the Köhler illumination), the 360° rotatable polarizer (for the production of polarized light), centering implements for the objectives and the analyzer attachment (for the analysis of the polarized light). The analyzer attachment is meant for receiving the 360° rotatable analyzer and optional available sliders (Bertrand lens for conoscopy; quartz wedge, lambda plate and lambda/4 plate used in order to get special colour effects). The wide field eyepieces (WF10x), which are also optional available, are placed at the trinocular head with an inclination of 30°. They have eye relief adjustment as well as diopter adjustment. The images shown by them are bright, true colored, sharp and rich in details. Apart from the slips and eyepieces, which were mentioned above, the accessories of the MPO 401 contain a dividing eyepiece (WF10x), the Abbe condenser with iris diaphragm, special polarization objectives (4x, 10x, 40x retractable, 100x-oil retractable), filters (blue, ground glass) and immersion oil. Nexcope NIB630 inverted research microscope with tiltable lighting unit
The NIB 630 is the flagship of the NIB600 series of inverted microscopes. With such an inverted design, the device enables the microscopy of plankton, microalgae and cell cultures. Cell cultures can be examined directly in the culture vessel without taking samples. For particularly easy sample handling, it is equipped with a tiltable lighting unit. This means that even tall vessels can be placed comfortably without bumping against the lighting unit. The microscope head is equipped with a height-adjustable ergo viewing head. This allows users of different heights to adjust the microscope to a comfortable and ergonomic viewing height with a single hand movement. This special microscope is used in water management offices, research institutes and universities, in human and veterinary medicine, agriculture, marine biology, limnology, aquaculture and many more. The integrated display informs you at any time about the selected magnification and the status of the illumination as well as the ECO mode. The brightness can be programmed for each objective so that there is no need to readjust when changing magnification and the user is not blinded. OBJECTIVE The NEXCOPE NIB630 microscope is equipped with a quintuple nosepiece. A 4x planachromatic objective and one each of 10x, 20x and 40x planachromatic phase contrast objectives are included. The condenser has a numerical aperture of 0.3 and an extra long working distance. The objectives are calculated for a soil thickness of 1.2 mm. Please use appropriate culture vessels. The universal specimen holder is suitable for standard slides. EYEPIECES The eyepieces have a field of view of 22 mm. This gives you a very large field of view. The high eye point makes them very comfortable even for spectacle wearers. A rubber edge protects against scratches on contact. Dioptric compensation from -5 to +5 can be individually adjusted for each eyepiece. The interpupillary distance can be adjusted between 48 mm and 75 mm. The binocular can be tilted up and down to adjust the viewing height. ILLUMINATION The Koehler illumination provides the best possible light guidance of the microscopy light. This avoids stray light and only illuminates the just visible area in the specimen. This protects sensitive living specimens from excessive light irradiation. The powerful 3W LED can be adjusted in brightness and ensures optimal illumination. The brightness can be adjusted individually for each objective. If an objective is changed, the NEXCOPE NIB630 automatically adjusts the previously set brightness. The display shows the currently selected lens and the current brightness. The tiltable lighting unit makes it easier to work with taller sample vessels. The adjustable condenser with extra-long working distance enables quick readiness for use. MECHANICS In addition to coarse focusing, fine focusing helps to bring the specimen into focus very precisely. A full turn of the fine focusing knob moves the stage up or down by 0.2 mm. On the right side, the fine focus can be adjusted with a finger. The friction of the coarse focusing can also be varied. The fine focusing knobs are held magnetically and can be mounted on the left or right. The XY stage offers the possibility of inserting various holders. In the microscope's basis, there are a transmitted light LED illuminator (3 W) as well as the power supply and the continuous adjustable dimmer for transmitted light. The trinocular head allows to mount a SLR camera or a USB microscope camera with an optional adapter. The inclination of the eyepiece tubes is 30°. The fluorescence attachment includes a special LED wheel with four different lamps (green, blue, violett, ultraviolett), a filter wheel and a radiation shield. The LED lamps produces the exciting radiation in order to get fluorescence. In contrast to usual light, polarized light has a preferred vibration plane of the light waves. Polarized light lets be seen anisotropic objects, whose observable properties are depending on the direction, "in a new light": crystals, but also animal hairs, feathers, muscles, nerve fibres and vegetable cell walls, which have a preferred direction of its biomolecules, show structures now, which were hidden in the normal bright field image. The 3D image also makes working with tools (e.g. preparing, dissecting, soldering) much easier.The zoom allows stepless adjustment of magnification with a zoom factor of 4.0 (1x-4x). Only minimal refocusing is needed when zooming. For this, the Advance ICD has a coaxial focus drive with coarse and fine adjustment. The screen delivers brilliant images without artefacts, as the display has the same resolution as the camera's output image. With the set of MikroCam and HDMI display, almost any trinocular microscope with camera connection can be upgraded to a digital microscope. This allows observation independently of a PC. The size of the display is optimally chosen so that the microscope user can microscope as usual and simultaneously view the image on the monitor. The display provides optimal image quality with a wide viewing angle of 178°! This allows several people to view the image without loss of quality. The microscope camera image is displayed on the HDMI screen always without delay and with a high refresh rate of 60 FPS. Displaying via HDMI microscope camera on an HDMI monitor is therefore usually of better quality than using a USB camera on a laptop or PC. This is particularly advantageous when working under microscopic control. Many users also benefit from improved ergonomics when viewing the image on the HDMI display. Especially higher magnifications with their high numerical aperture are uncomfortable for eye problems such as floaters (vitreous opacities). Digital viewing can provide relief here. The display can be powered with the included Y-cable using the same power supply as the camera. A suitable, short, angled HDMI cable is also included to avoid cable clutter at the workplace. Mounting on a MikroCam PRO or MINI is done using two screws. Screws and tools are included.
