Typische Anwendungen für die Dunkelfeldmikroskopie

Das Dunkelfeld eignet sich als Kontrastverfahren immer dann besonders, wenn transparente Zellen oder Mikroorganismen sichtbar gemacht werden sollen, die selbst wenig Kontrast haben. Typische Beispiele sind zB Amöben oder Epithelzellen, zB von der Mundschleimhaut. Hier kann dann auf eine Fixierung und anschließene Färbung verzichtet werden. Dunkelfeldmikroskopie eignet sich auch besonders für Lebendkulturen von Wasserorganismen oder Algen, die man ja in Bewegung sehen möchte, was eine Färbung meistens ausschließt. Zusätzlich zum erzielten Kontrastgewinn sind Dunkelfeldbeobachtungen auch ästhetisch besonders reizvoll!

Schiefe Beleuchtung

Bei der schiefen Beleuchtung wird eine Kontrastierung erzielt, indem ein Teil bzw. eine Hälfte des normalen Beleuchtungsstrahlengangs abgedeckt wird. Kommerzielle Lösungen für dieses Kontrastverfahren sind leider selten, man kann diesen Effekt aber mit einer selbst angefertigten Blende / Schieber in der Filterebene des Kondensors erzielen. Oft kann auch ein vorhandener Filterhalter oder eine Hilfslinse am Kondensor teilweise in den Strahlengang eingeschwenkt werden. Der Rahmen des Filterhalters bzw. der Feldlinse blockiert dann einen Teil des Lichts. Das Präparat wird dann schräg durchstrahlt, es ergibt sich quasi ein Schattenwurf an Feinstrukturen des Präparats.

Phasenkontrast

Beim Phasenkontrastverfahren wird eine Kombination aus einem speziellen Kondensor mit wechselbaren Blenden (diese erzeugen einen Lichtring) und entsprechenden Phasenkontrastobjektiven (mit Phasenring) verwendet. Das Licht, welches das Präparat durchstrahlt, wird dort gebeugt und erfährt an Bestandteilen mit unterschiedlichem Brechungsindex eine Phasenverschiebung. Licht, welches keine Beugung erfährt, wird am Phasenring geblockt. 

Ãœblicherweise werden separate Phasenkontrastsätze angeboten. Eine ausschließliche Ausstattung des Mikroskops nur mit Phasenkontrastobjektiven ist in der Regel nicht empfehlenswert, weil zwar die Auflösung der Objektive durch den Phasenring nicht beeinträchtigt wird, der Kontrast aber schlechter ist als bei normalen Hellfeldobjektiven. Auch für Polarisation eignen sich diese Objektive nicht. 

Ein Phasenkontrastkondensor muss sorgfältig zentriert werden, damit Lichtring und Phasenring aufeinander abgebildet werden. Daher ist ein Wechsel zwischen Phasenkontrastkondensor und Hell-oder Dunkelfeldkondensor aufwändig. Man kann aber einzelne Hellfeldobjektive im Revolver neben mit Phasenobjektiven verwenden, denn der Phasenkontrastkondensor hat einen Durchlass für Hellfeld. Für die Zentrierung bzw. die Höheneinstellung des Kondensors ist im Phasenkontrastsatz ein Hilfsokular (auch "Einstellfernrohr" genannt) enthalten, so dass auf die Ebenen von Phasen- und Lichtring scharfgestellt werden kann.

Die Bedingungen für Phasenkontrast sind optimal nur für grünes Licht erfüllt. Daher ist ein entsprechendes Grünfilter hilfreich. Da ohnehin farblose Objekte im Phasenkontrast beobachtet werden, ist eine Einfärbung des Bildes tolerierbar.

Typische Anwendungen für Phasenkontrast

Ähnlich wie beim Dunkelfeld eignet sich Phasenkontrast für Objekte, die farblos sind und wenig Kontrast haben. Darunter fallen z.B. Planktonorganismen oder Belebtschlamm. Auch Zellkulturen oder Zellen im Urinsediment können mit Phasenkontrast besser dargestellt und damit auch schneller und sicherer beurteilt werden als im Hellfeld. Auch die Zählung von Asbestfasern erfolgt bei einigen Standardprotokollen im Phasenkontrast.

Polarisation

Durch Polarisation können optisch aktive  bzw. doppelbrechende Präparatstrukturen besonders hervorgehoben werden. Das können zB Minerale sein, aber auch viele Kunststoffe, Naturstoffe wie Stärke etc. zeigen diesen Effekt. Weniger bekannt ist, dass man zB auch an lebenden Organismen interessante Strukturen hervorheben kann, so zB Muskelfasern von Daphnien oder Rädertierchen. Die Polarisation dient in der Industrie vor allem zur Charakterisierung von Materialien. Ist die Schichtdicke eines Materials bekannt, so kann anhand der entstehenden Interferenzfarben auch auf das Material geschlossen werden. In der Materialforschung wird die Polarisation zB auch zur Untersuchung von Spannungen im Material durch Spannungsdoppelbrechung genutzt. Besonders Spritzgussteile oder Kunststofffolien bzw. Fasern können so auf Herstellungsfehler untersucht werden. Die klassische Anwendung ist natürlich die Geologie / Mineralogie, bei der Gesteinsdünnschliffe in polarisiertem Licht untersucht werden.

Aufgrund der Spektralverteilung sind in der Polarisationsmikroskopie Halogenbeleuchtungen besser geeignet als LED.

In contrast to usual light, polarized light has a preferred vibration plane of the light waves. Polarized light lets be seen anisotropic objects, whose observable properties are depending on the direction, "in a new light": crystals, but also animal hairs, feathers, muscles, nerve fibres and vegetable cell walls, which have a preferred direction of its biomolecules, show structures now, which were hidden in the normal bright field image. The NIB 630 is the flagship of the NIB600 series of inverted microscopes. With such an inverted design, the device enables the microscopy of plankton, microalgae and cell cultures. The BRESSER DST-0745 is a special microscope that is operated with a c-mount camera (camera not included). This allows direct viewing on a screen. Due to the vertically installed optics, there is no shifting of the image when focusing. This is especially useful for "focus stacking" shots. The zoom is continuously adjustable from 0.7x - 4.5x. A coarse and fine focus adjustment facilitates focusing, especially at high magnifications. The convenient and user-friendly operation makes the digital microscope particularly suitable for daily use in the workshop. Measurements, quality control of surfaces or inspection of components and raw materials are easy with the workshop microscope. Insects, plants and rocks are also impressively displayed.
