Bei den professionellen Mikroskopen verfügt der Beleuchtungsstrahlengang über eine Leuchtfeldblende, einen höhenverstellbaren und zentrierbaren Kondensor (Abbe-Kondensor) und eine Kondensorblende. Hiermit kann der Strahlengang optimal an das Objektiv und dessen numerische Apertur (der Öffnungswinkel des Objektivs) angepasst werden. Dies nennt man Köhlersche Beleuchtung. Bei höherer Vergrößerung (höhere Apertur des Objektivs) muss die Kondensorblende weiter geöffnet werden. Auch wenn die Frontlinse der Objektive bei höherer Vergrößerung immer kleiner wird, so wird doch dieser Effekt durch den viel kleineren Arbeitsabstand überkompensiert. Die Frontlinse sitzt sehr viel näher an der Probe und damit wird ein größerer Öffnungswinkel notwendig, um das Objektiv komplett auszuleuchten.

The BRESSER Science MTL 201 is a special reflected light microscope, which allows to observe non-transparent surfaces at higher magnifications (50x-800x). Tiefenschärfe

Bei der Einstellung der Köhlerschen Beleuchtung ist das Bildergebnis immer ein Kompromiss aus Auflösung, Kontrast und Tiefenschärfe, wie im Beispielbild zu sehen. Bei zu starkem Abblenden (Tiefenschärfe hoch) gehen feine Details der Probe verloren. Daher arbeitet man für hochauflösende Ergebnisse eher mit Fokus-Stacking, d. h. man nimmt mehrere Bilder mit unterschiedlichen Fokusebenen auf und mittelt diese anschließend. Unsere MikroCams verfügen in der Software über eine entsprechende Stacking-Routine. Dies kann bei einigen Modellen bereits im Livebild, d. h. während die Probe durchfokussiert wird, erfolgen.

Einfache Mikroskope verfügen meistens nur über eine simple Kondensorlinse im Tisch und sind damit auf eine Arbeitsblende festgelegt, egal welches Objektiv verwendet wird. Die maximale Auflösung der höher vergrößernden Objektive wird damit nicht ganz erreicht, aber das Bild sieht für Einsteiger ansprechender aus (mehr Kontrast, mehr Tiefenschärfe) und man findet sich auch in der Probe besser zurecht. Das ist z. B. beim Durchmustern einer größeren Probenmenge bzw. einer höheren Schichtdicke hilfreich. Zudem muss am Mikroskop nichts eingestellt oder justiert werden.

In contrast to usual light, polarized light has a preferred vibration plane of the light waves. Polarized light lets be seen anisotropic objects, whose observable properties are depending on the direction, "in a new light": crystals, but also animal hairs, feathers, muscles, nerve fibres and vegetable cell walls, which have a preferred direction of its biomolecules, show structures now, which were hidden in the normal bright field image. Images made by polarized light are often characterized by a fantastic coloring. Polarization microscopes are used particularly in mineralogy, geology, biology and medicine. In order to change the object's direction in a controlled way, the Science MPO 401 has a round, center-adjustable and full rotatable object stage with degree scale.
